My role
- High-Throughput Screening and Assay Optimization:
- Performing high-throughput screenings of antisense oligonucleotides (ASOs) for GCaMP expression, neurite outgrowth, and cardiotoxicity using the ImageXpress Micro-confocal system and automated platforms such as the Integra Viaflo (96/384), Hamilton, and Echo.
- Developing pilot transfection experiments to ensure accurate ASO targeting within cells.
- Maintaining extensive cell culture processes for neurons and cardiomyocytes, and managing imaging, quality control, and analysis for GCaMP, neurite outgrowth, and cardiotoxicity.
- Writing Standard Operating Procedures (SOPs) for assays and experimental setups to standardize laboratory operations.
- Laboratory Automation and Custom Programming:
- Doing the automation of laboratory processes by programming systems including the Hamilton, Echo, and Integra Viaflo for efficient drug dilution and assay preparation.
- Designing custom scripts for automating assays which improve efficiency and reproducibility.
- Molecular Biology and Advanced Sequencing Techniques:
- Enhancing ASO detection with padlock probes and conducting analyses using Leica systems.
- Implementing the Targeted Allele Counting Sequencing (tacseq) method on the Aviti sequencer, and optimizing the protocol for cost-effectiveness and automation compatibility.
- Data Analysis and Software Development:
- Developing a Python-based analysis pipeline for high-throughput cardiotoxicity data that assigns hazard scores to drugs.
- Writing a script for single cell neurotoxicity data analysis which does ROI detection, individual peak calculation, baseline correction, and activity comparison.
- Creating Python scripts to automate steps like configuration file creation and image optimization to streamline workflows.
- Stem Cell Research and iPSC Differentiation:
- Assisting in the differentiation of iPSCs to develop a cost-effective neuro-model for ASO screenings, using advanced cell culture techniques.
